Effects of catgut embedding at stellate ganglion on lung tissue injury and HPA axis in asthmatic rats

Abstract

Objective It is to investigate the effects of catgut embedding at stellate ganglion on the symptoms,lung tis-sue injury,and hypothalamic-pituitary-adrenal(HPA)axis in rats with bronchial asthma.Methods Forty SPF-grade male SD rats were randomly divided into 5 groups:control group,model group,catgut embedding group,block group and hor-mone group,with 8 rats in each group.The rats of all groups except for the control group were sensitized and stimulated with ovalbumin to establish asthma models.Starting from the stimulating experiment,the catgut embedding group was given catgut embedding at stellate ganglion once;the block group was given stellate ganglion block,the hormone group was given budesonide nebulization,all administered once daily,continuously treated for 7 days;the control group and model group were not treated.The general condition of the rats was observed,and their body weight was recorded;the changes of lung morphology of the rats were observed by HE staining and Masson's trichrome staining,the inflammatory cell infiltration scores and collagen fiber deposition degrees were calculated;the positive expression of substance P(SP)in lung tissue of the rats were detected by immunohistochemistry;the contents of corticotropin-releasing hormone(CRH),adrenocortico-tropic hormone(ACTH),and corticosterone(CORT)in serum and alveolar lavage fluid of the rats were measured by ELISA.Results Compared with the blank group,the rats in the model group developed symptoms such as wheezing,short-ness of breath and restlessness,and their body weights were significantly reduced(all P<0.05);the pathological injury in lung tissue was obvious,the inflammatory cell infiltration scores and collagen fiber deposition degrees were significantly in-creased(all P<0.05);the mean optical density value of SP positive expression in lung tissue was significantly increased(P<0.05),while the contents ofCRH,ACTH,and CORT in serum and alveolar lavage fluid were significantly reduced(all P<0.05).Compared with the model group,the symptoms such as wheezing,shortness of breath and restlessness of the rats in the catgut embedding group,block group and hormone group were improved,and their body weight were signifi-cantly increased(all P<0.05);the pathological injuries in lung tissue were significantly alleviated,the inflammatory cell infiltration scores and collagen fiber deposition degrees were significantly decreased(all P<0.05);the mean optical densi-ty values of SP positive expression in lung tissue were significantly decreased(all P<0.05),while the contents of CRH,ACTH,and CORT in serum and alveolar lavage fluid were significantly increased(all P<0.05).Conclusion Catgut en-bedding at stellate ganglion can significantly alleviate the symptoms and improve the lung tissue injury of asthmatic rats.The possible mechanism of action may be related to the down-regulation of SP expression and the improvement of HPA axis function.
