响应面优化棉花茎杆皮色素提取工艺及抗氧化研究
Study on Response Surface Methodology Optimization of Pigment Extraction from Cotton Stem Bark and Its Antioxidant Activity

Abstract
To optimize the extraction process of natural pigments from cotton stalk fibers and analyze their antioxidant activity,the 1,1-diphenyl-2-picrylhydrazyl(DPPH)radical scavenging activity,2,2'-azino-bis 3-ethylbenzothiazoline-6-sulfonic acid(ABTS)radical scavenging ability and iron-reducing power were taken as the key indicators.A strategy combining ultrasonic-assisted technology and response surface methodology was employed to optimize the extraction conditions.The results showed that the optimal pigment extraction conditions were as follows:liquid-to-material ratio of 104.036 mL·g-1,ethanol content of 73.462%,ultrasonic temperature of 59.801℃,and extraction time of 100.056 min.Under these conditions,the extracted pigment content was 51.646 mg·g-1.The antioxidant test indicated that this pigment had a strong scavenging ability for DPPH,ABTS free radicals and iron ions,and the half-maximal inhibitory concentration(IC50)for DPPH was 46.201 39 µg·mL-1,while for ABTS the IC50 was 19.542 30 µg·mL-1.The iron-reducing assay test showed that at a pigment content of 1 mg·mL-1,its activity was almost equivalent to that of vitamin C.Above results opened new avenues for the comprehensive utilization of cotton,particularly in resource-efficient applications and food additive development,showing great potential for future application.关键词
