Abstract

This study elucidated the role of GSDMD mediated pyroptosis in attenuated Mycobacterium bovis strain BCG infection.Macrophages were infected with BCG(MOI=10),and cells were collected at various time points for detection of protein levels of NLRP3,cleaved caspase-1,and GSDMD-N.The formation of ASC specks was also detected.qPCR and ELISA were used to deter-mine IL-1β mRNA and protein levels after BCG infection.Finally,the percentage cell death in BCG-infected bone marrow-derived macrophages from GSDMD knockout mice was detected with pyridine iodide staining.The attenuated M.bovis strain BCG induced a significant increase in NLRP3 protein levels after infecting macrophages.NLRP3 formed a platform by binding ASC,and subsequently recruited pro-caspase-1 and activated caspase-1.Consequently,BCG activated GSDMD and increased IL-1β secretion,thus ulti-mately inducing pyroptosis,a critical process in BCG clearance.This study demonstrated that BCG infection induces GSDMD-mediated pyroptosis in mouse macrophages,and this pyroptosis restricts BCG survival.The results provide a theoretical basis for fur-ther understanding of the mechanism of BCG-induced protective immunity and modification of the BCG vaccine.
