Ca2+/cAMP mediates activation of NLRP3 inflammatory bodies in ECA109 cells infected with Newcastle disease virus

Abstract
This study investigated the associations among activation of the calcium-sensing receptor(CASR),intracellular pH changes,and NLRP3 inflammasome activation in ECA109 esophageal cancer cells after Newcastle disease virus(NDV)infection.An additional aim was to determine whether alterations in intracellular Ca2+and cyclic adenosine monophosphate(cAMP)levels mediated by CASR activation might play critical roles in the NLRP3 inflammasome activation process.Intracellular Ca2+concentrations were de-tected with fluorescence probe technology;IL-1β levels in the supernatant were measured with ELISA;the pH of the cell culture me-dium was detected with a pH meter;ultrasonic extraction of the cytoplasm combined with ELISA was used to detect the levels of inosi-tol trisphosphate(IP3)and cAMP;western blotting was used to detect the protein expression of CASR,NLRP3,and IL-1β;and im-munofluorescence technology was used to detect the formation of apoptosis associated spot like protein containing a CARD(ASC).Af-ter infection with NDV F3,the NLRP3 inflammasome was activated,and CASR protein expression was upregulated.The pH of the cell culture medium decreased,and the concentration of intracellular Ca2+significantly increased(P<0.05).Moreover,the IP3 content in-creased,whereas the cAMP content decreased(P<0.01).After NDV F3 infection of ECA109 cells,the NLRP3 inflammasome was ac-tivated.The mechanism of activation might potentially involve disruption of the intracellular acid-base balance after viral infection,thus leading to activation of the Calcium-Sensing Receptor(CASR).The increase in intracellular Ca2+levels and the decrease in cAMP levels after CASR activation are likely to be involved in NLRP3 inflammasome activation in NDV F3-infected ECA109 cells.关键词
