改良抗酸菌荧光染色镜检试剂的研究
Modification of reagents for fluorescence staining microscopic examination of acid-fast bacilli

Abstract
To decrease the use of hazardous chemicals,this study reports an evaluation of the staining effects of using Triton X-100-Vaseline instead of phenol,using methylene blue instead of potassium permanganate,and reduced amounts of alcohol and hydro-chloric acid to prepare a decolorization solution for acid fast bacterial fluorescence staining.Sputum samples were collected from pa-tients with suspected pulmonary tuberculosis.Smear staining microscopic examination and MGIT960 fluid culture were performed ac-cording to standard tuberculosis laboratory testing procedures.A Triton X-100-Vaseline water suspension was used instead of a phe-nol water suspension to prepare the initial staining solution(auramine O working solution),potassium permanganate counterstaining solution was replaced with 0.1%methylene blue,and 5%hydrochloric acid(95%alcohol)was replaced with 0.5%hydrochloric acid(75%alcohol)as the decolorization solution.After staining and microscopic examination,the sensitivity and specificity of the new re-agents were compared with the standard staining method according to culture results.The standard staining results had a sensitivity and specificity of 57.14%(40/70)and 98.69%(151/153),respectively.The sensitivity and specificity of Triton X-100-Vaseline were 55.71%(39/70)and 98.04%(150/153),respectively.The sensitivity and specificity of the 0.5%hydrochloric acid 75%alcohol method were 57.14%(40/70)and 98.04%(150/153),respectively.The sensitivity and specificity of the 0.1%methylene blue method were 55.71%(39/70)and 98.04%(150/153),respectively,No significant differences in the results were observed for the three modi-fied reagents versus the standard method(P>0.05).On the basis of the evaluation data,in acid fast fluorescence staining,Triton X-100-Vaseline can replace phenol in preparation of the initial staining solution.In the decolorization solution,0.5%hydrochloric acid-75%alcohol can be used,and 0.1%methylene blue aqueous solution can be used as a potassium permanganate substitute for counter-staining.关键词
