口蹄疫病毒3D蛋白与宿主互作蛋白的筛选及鉴定
Screening and identification of host proteins interacting with 3D protein of foot-and-mouth disease virus

Abstract
Foot-and-mouth disease(FMD)is an acute,febrile,and highly contagious infectious dis-ease of cloven-hoofed animals such as pigs,cattle,and sheep,caused by foot-and-mouth disease virus(FMDV).The FMDV 3D protein is the RNA-dependent RNA polymerase(3Dpol)encoded by the virus,which is crucial for viral RNA replication.However,there are few reports on the interaction between 3D and host proteins.In this study,a yeast two-hybrid system was used with a cDNA yeast expression library from porcine kidney epithelial PK-15 cells,and 15 potential host proteins interacting with 3D were ulti-mately obtained.Further validation by co-immunoprecipitation and confocal microscopy identified that host ataxin-3 and VDAC1 interact with 3D.The findings provide data support for the role of host protein-3D interactions in viral replication.关键词
