Abstract

One"cartridge-based"rapid nucleic acid detection kit for African swine fever virus(ASFV)is described in this paper,the kit integrates nucleic acid extraction reagents and fluorescent PCR reaction solutions into a fully enclosed"cartridge-based"reaction tube,enabling automated com-pletion of nucleic acid extraction and PCR amplification within 25 minutes.Performance evaluation demonstrated 100%specificity and a sensitivity of up to 608.5 copies/mL.The intra-and inter-assay coefficients of variation(CV)for medium and low concentration nucleic acid samples were both below 5%,indicating excellent repeatability and reproducibility.Validation with 90 known samples showed that,compared to the WOAH-recommended fluorescent PCR method(1.3.4 protocol),the kit achieved the concordance rates of 100%(41/41)for positive samples and 97.96%(48/49)for negative samples,with one negative sample tested to be positive with the cartridge-based kit,resulting in an overall concor-dance rate of 98.89%(89/90).Among 87 clinical samples of unknown infection status randomly collected from the market,the cartridge-based kit screened 14 positive samples,all of which were negative by the WOAH-recommended PCR.These findings suggest that the"cartridge-based"kit offers a superior tool for on-site rapid detection of ASFV with higher sensitivity and easily used.
