Establishment and application of PCR detection method based on Anaplasma marginale msp4 gene

Abstract
To establish a specific,sensitive and rapid PCR assay for Anaplasma marginale,a primer pair targeting the conserved region of the msp4 gene was designed and the reaction conditions were opti-mized.The assay was then evaluated for sensitivity,specificity and repeatability,and applied to clini-cal samples.The method exclusively detected A.marginale DNA and showed no cross-reactivity with seven common blood-borne protozoa of domestic animals.The detection limit was 380 fg/μL(≈126 copies per re-action).Both intra-assay and inter-assay results indicated the method had high repeatability and sta-bility.Testing of 120 clinical samples revealed a positivity rate of 54.1%(65/120),significantly higher than the 37.5%(45/120)obtained with a previously reported PCR.In conclusion,the newly devel-oped PCR provides a sensitive,specific and rapid tool for the epidemiological surveillance and accurate diagnosis of bovine anaplasmosis in China.关键词
