Optimization of the Extraction of Citrus Peel Flavonoids and Their Antioxidant Activity by Response Surface Methodology

Abstract

The citrus varieties of the highest hesperidin and naringin contents were screened out from 15 citrus species in the resource nursery,and the optimization of the extraction process and the study on antioxidant activity were carried out.The results showed that the varieties with the highest contents of hesperidin and naringin were Mihaya and grapefruit,respectively.The optimal conditions for the extraction of hesperidin and naringin were as follows:for hesperidin,solid-liquid ratio of 1∶36.19(g/mL),ultrasonic temperature of 45.20℃,and ultrasonic time of 47.47 min;for naringin,solid-liquid ratio of 1∶43.32(g/mL),ultrasonic temperature of 53.48℃,and ultrasonic time of 53.47 min.The predicted values of the yield of hesperidin and naringin were 12.75%and 8.31%,respectively.The antioxidant studies showed that the IC50 of scavenging DPPH radicals were 0.52 mg/mL and 5.57 mg/mL for hesperidin and naringin,respectively,while that of scavenging ABTS radicals were 0.41 mg/mL and 3.78 mg/mL.Moreover,0.8 mg/mL of hesperidin and 4.72 mg/mL of naringin in the ability to reduce Fe3+were roughly equivalent to 0.6 mg/mL of vitamin C.This study results in an optimal protocol for the extraction of hesperidin and naringin,and the antioxidant activity of hesperidin is higher than that of naringin.
