基于网络药理学与分子对接探究天葵子与预知子抗肝癌作用机制
Exploration on the Mechanism of Semiaquilegiae Radix and Akebiae Frucyus against Liver Cancer Based on Network Pharmacology and Molecular Docking

Abstract
Objective To investigate the targets and potential molecular mechanisms of the combination therapy of Semiaquilegiae Radix and Akebiae Frucyus in the treatment of liver cancer.Methods The active components and the targets of Semiaquilegiae Radix and Akebiae Frucyus were obtained from the TCMSP and SwissTargetPrediction databases.Liver cancer targets were collected through GeneCards and OMIM databases.The drug-component-target-disease network was constructed by Cytoscape 3.10.2.Based on the STRING database,the protein interaction(PPI)network of the co-targets of drugs and liver cancer was established,and the core genes were screened.Target gene enrichment analysis was further performed,and the binding ability of core genes and active components was tested by molecular docking technology.Results After screening,7 and 6 active components contained in Semiaquilegiae Radix and Akebiae Frucyus were obtained,corresponding to 402 and 306 potential targets,respectively.6 838 targets related to liver cancer were obtained,and the core targets included HIF1A,CCND1,ESR1,CASP3 and HSP90AA1.Enrichment analysis showed that the signaling pathways involved in Semiaquilegiae Radix and Akebiae Frucyus in the treatment of liver cancer were mainly cancer-related pathway,apoptosis pathway,tumor proteoglycan metabolism pathway,Rap1 signaling pathway and Ras signaling pathway.Molecular docking analysis showed that the main active components of Semiaquilegiae Radix and Akebiae Frucyus showed strong binding activity with key target proteins of liver cancer.Conclusion Semiaquilegiae Radix-Akebiae Frucyus exerts a synergistic anti-liver cancer effect by regulating the key targets HIF1A,CCND1 and HSP90AA1 through various active components,including berberrubine,β-sitosterol and sitosterol.关键词
