Optimization of extraction process for polysaccharides from Lyophyllum decastes by orthogonal test and their antioxidant activity in vitro

Abstract

For the efficient preparation of Lyophyllum decastes polysaccharides(LDPs),this study systematically compared the effects of five extraction methods—hot water extraction(HWE),enzyme-assisted aqueous extraction with alcohol precipitation(EAAE),microwave-assisted water extraction(MAE),ultrasound-assisted water extraction(UAE),and freeze-thaw-assisted water extraction(FTE)—on the extraction yield and polysaccharide content.The results showed the following respective yields and polysaccharide contents:HWE:(4.63±0.40)%and(41.87±1.39)%;EAAE:(19.38±1.90)%and(77.03±1.76)%;MAE:(7.32±0.47)%and(44.78±0.76)%;UAE:(8.57±0.48)%and(42.57±1.36)%;FTE:(11.46±0.50)%and(54.59±2.22)%.Among these,EAAE demonstrated the best performance and was therefore selected for LDPs extraction.Subsequently,based on single-factor experiments,the extraction process was optimized using an orthogonal array design focusing on four key parameters:extraction cycles,enzymatic hydrolysis time,enzyme concentration,and solid-liquid ratio.The optimal conditions were determined as follows:3 extraction cycles,150 min enzymatic hydrolysis time,2.5%enzyme concentration,and a solid-liquid ratio of 1∶20(g∶mL).Under these optimized conditions,triple validation experiments confirmed the process's stability and reliability,showing polysaccharide contents of 83.19%,82.08%,and 81.96%,and extraction yields of 20.18%,21.34%,and 20.96%,respectively.Furthermore,in vitro antioxidant activity assays revealed that the prepared LDPs exhibited potent scavenging abilities against both DPPH·and ABTS+·in a dose-dependent manner.At a concentration of 4.0 mg/mL,the scavenging rates reached 80.17%for DPPH·and 88.42%for ABTS+·.In conclusion,this study successfully established an efficient extraction process for LDPs and confirmed their significant antioxidant potential,providing a crucial foundation for the development and application of Lyophyllum decastes in functional foods.
