响应面法优选凤眼草口服液提取工艺研究
Optimization of extraction technology for fruits of Ailanthus altissima(Mill.)Swingle oral liquid by response surface methodology

Abstract
The objective was to optimize the water extraction process parameters of Ailanthus altissima(Mill.)Swingle oral liquid and determine the optimal extraction conditions.The overall desirability(OD)value,calculated from quercetin and kaempferol contents as well as extract yield,was used as the comprehensive evaluation index.Single-factor experiments were conducted to investigate the effects of solid-liquid ratio,extraction time,and extraction frequency on the extraction efficiency,thereby determining the appropriate range for each factor.Based on these results,the Box-Behnken central composite design principle was applied,and response surface methodology was employed to systematically optimize the water extraction process.Response surface analysis demonstrated that the established mathematical model had a coefficient of determination(R²)of 0.9903 and an adjusted coefficient of determination(R²Adj)of 0.9779.The close agreement between these two values indicated excellent model fitting.Analysis of variance showed that the model was highly significant(P<0.01),while the lack-of-fit was not significant(P>0.05),further confirming the reliability of the model.The optimal water extraction conditions were determined as follows:solid-liquid ratio of 1∶17(g∶mL),extraction time of 2 h,and extraction frequency of 3 times.Under these optimized conditions,the quercetin content was 1.07 mg/mL,kaempferol content was 0.17 mg/mL,extract yield was 25.65%,with a mean OD value of 0.940 and RSD of 1.2%.The water extraction process of Ailanthus altissima oral liquid optimized by response surface methodology is stable and feasible,with scientifically sound process parameters.This study provides a foundation for the further development and industrial production of Ailanthus altissima preparations.关键词
