摘要

Abstract

YTH proteins are a class of proteins containing the YTH domain,which can specifically bind RNA,participate in N6-methyladenosine(m6A)modification,and play a key role in biotic and abiotic stress responses.Preliminary work in this study identified 5 drought-responsive YTH genes via transcriptome sequencing.We identi-fied 21 YTH domain-containing genes based on the HMMER model using the potato variety of'Favorita'as the ex-perimental material to elucidate their drought response mechanisms.By integrating bioinformatics analyses(chromo-somal localization,gene structure,phylogenetic relationships,and cis-element prediction)with expression validation under drought stress,their features and expression patterns were characterized.Additionally,protein-pro-tein interaction(PPI)networks were constructed using the STRING database to reveal functional associations.The results showed that these YTH genes were distributed across 5 chromosomes.Among them,the 5 drought-responsive genes were identified as StYTH1.1,StYTH1.3,StYTH1.4,StYTH1.5,and StYTH2.1.Furthermore,structural and conserved motif losses were observed in StYTH3.1 and StYTH4.1.Phylogenetic tree and gene structure analyses re-vealed that the potato YTH family could be classified into 3 subfamilies,and collinearity analysis indicated that gene duplication events drove the evolution of the potato YTH family.The phylogenetic tree showed that potato YTH genes were more closely related to those of tomato.Additionally,drought-inducible MYB binding sites and multiple plant hormone-related functional elements were detected in StYTH 1.1/1.2/1.3/1.4/1.5/3.1/6.1/6.2/8.1.The STRING database predicted 20 partner proteins for StYTH proteins,which,together with the 5 drought-responsive StYTH proteins,formed a complex protein interaction network primarily involved in regulating mRNA splicing modification,biotic and abiotic stress responses,and signaling pathways mediated by abscisic acid(ABA),sali-cylic acid(SA),and other plant hormones.
