STAT3和FGL1在乳腺癌中表达的临床意义及预后价值
Clinical significance and prognostic value of STAT3 and FGL1 expression in breast cancer

Abstract
Objective:Signal transducer and activator of transcription 3(STAT3)and fibrinogen-like protein 1(FGL1)are closely associated with tumor initiation and progression,yet studies on the clinical significance of their combined detection in breast cancer are still lacking.This study aims to explore the clinical significance and prognostic value of STAT3 and FGL1 expression in breast cancer. Methods:A total of 208 paraffin-embedded breast cancer tissues from female patients and 208 adjacent non-cancerous tissues archived in the Department of Pathology,Henan Chest Hospital,from January 2016 to December 2018 were retrospectively collected.Immunohistochemistry was used to detect STAT3 and FGL1 protein expression in breast cancer and adjacent tissues.Differences in expression between breast cancer and adjacent tissues,among molecular subtypes(luminal,ERBB2-positive,and basal-like),correlations between STAT3/FGL1 expression and clinicopathological parameters,prognostic factors of breast cancer,and the relationship between STAT3/FGL1 expression and patient survival were further analyzed. Results:Compared with adjacent tissues,the positive expression rates of STAT3 and FGL1 in breast cancer tissues were significantly increased(both P<0.05).STAT3 positivity showed no statistically significant difference among the three molecular subtypes(P=0.169).FGL1 positivity differed significantly among the subtypes(P=0.006),with ERBB2-positive and basal-like subtypes showing significantly higher positive rates than the luminal subtype(P=0.011 and P=0.006,respectively),while no significant difference was observed between ERBB2-positive and basal-like subtypes(P=0.755).STAT3 positivity was associated with tumor diameter,histological grade,lymph node metastasis,and PR status(all P<0.05).FGL1 positivity was associated with patient age,tumor diameter,histological grade,lymph node metastasis,ER status,and PR status(all P<0.05).STAT3 and FGL1 expression were significantly higher in histological grade Ⅲ than in grade Ⅱ(both P<0.001),but comparisons between grade Ⅲ or grade Ⅱ versus grade Ⅰ showed no statistically significant differences(both P<0.016 7,adjusted significance level was set at α'=0.0167).Univariate and multivariate analyses showed tumor diameter and STAT3 expression status as independent risk factors for poor prognosis in breast cancer(both P<0.05).Patients with positive STAT3 or FGL1 expression had significantly lower 5-year overall survival rates than those with negative expression(both P<0.05). Conclusion:STAT3 and FGL1 are highly expressed in breast cancer tissues.STAT3 positivity is an independent risk factor for poor prognosis in breast cancer patients,and although FGL1 is not an independent prognostic risk factor,it is one of the factors affecting 5-year overall survival.STAT3 and FGL1 may serve as potential biomarkers for clinical evaluation of disease condition and prognosis in breast cancer patients.关键词
