摘要

Abstract

Ischemic stroke(IS)is one of the leading causes of death and disability.Recent studies have shown that the pathological processes following a stroke are not only limited to blood flow obstruction,but also involve complex immune cell functions,such as the regulation of inflammatory responses and brain tissue repair and remodeling.The innate immune cells in the brain and peripheral immune cells play multiple roles at different stages of stroke.Although traditional research methods such as flow cytometry and transcriptome sequencing have revealed the basic information of immune responses,they are unable to deeply analyze the heterogeneity and spatial distribution of cells.The emergence of single-cell omics has provided new perspectives for studying post-stroke immune cell subpopulations,lineage differentiation trajectories,and intercellular interaction networks,has greatly promoted the in-depth understanding of the immune mechanism of stroke.This paper reviews the application progress of single-cell omics technologies in the study of the immune microenvironment in ischemic stroke,focusing on elaborating its research achievements in immune cell recognition and heterogeneity analysis,tracking dynamic changes in the immune microenvironment,intercellular interactions(including cross-lineage interactions),and the discovery of related therapeutic targets,also discusses the challenges in current research and the future development directions,which providing new references for the basic research and clinical translation of ischemic stroke.
