甘蓝型油菜冷调节基因BnaCOR27的克隆及表达分析
Cloning and Expression Analysis of BnaCOR27 Gene in Brassica napus

Abstract
The COR27 gene is a cold regulatory gene important in plant response to cold stress.To explore the function of COR27 gene from Brassica napus,the variety of ZS11 was used and a COR27 full-length CDS se-quence of 723 bp was cloned.,It encoded 240 amino acids and was named as BnaCOR27.Bioinformatics analy-sis showed that the BnaCOR27 protein was a hydrophilic protein with poor stability,had no transmembrane do-main and signal peptide,and was a non-secreted protein.Multiple sequence alignment revealed low sequence i-dentity between BnaCOR27 and COR27 proteins of other 17 species such as Brassica rapa,Hirschfeldia incana,and Raphanus sativus.The phylogenetic evolutionary tree showed that the COR27 proteins of Cruciferae were clustered together,and BnaCOR27 had the most close evolutionary relationship with COR27 of B.rapa.Promoter element analysis showed that the expression of BnaCOR27 was not only induced by low temperature but also reg-ulated by light signals,and it played an important role in response to cold stress as well as other stress stresses.The results of real-time fluorescence quantitative PCR detection showed that BnaCOR27 had the expression pattern of leaf﹥root﹥flower﹥stem﹥silique in B.napus ZS11.The overexpression vector pCAMBIA1301-BnaCOR27 was successfully constructed.These results could lay the foundation for in-depth exploration of the mechanism of BnaCOR27 gene in cold resistant process and provide a theoretical guidance for molecular breeding of B.napus.关键词
