陆地棉SCL3亚基因家族的鉴定及其在干旱胁迫应答中的功能研究
Identification of the SCL3 Subfamily in Gossypium hirsutum and Functional Analysis in Drought Stress Response

Abstract
To identify the gene members of SCL3,a key subfamily of the GRAS family in cotton,and clarify their functions in cotton(Gossypium hirsutum)drought resistance,in this study,SCL3 subfamily genes were systematically identified using bioinformatics methods,and their physicochemical properties,gene structures,cis-acting elements in the promoter region,tissue expression specificity,and expression patterns under drought stress were analyzed.Meanwhile,the function of GhSCL3-D13-1 in cotton response to drought stress was preliminarily verified by virus-induced gene silencing(VIGS)technology.The results showed that 10 GhSCL3 genes were identified from the cotton genome,and all their encoded proteins were unstable hydrophilic proteins.Conserved motif analysis indicated that genes with close phylogenetic relationships had highly similar conserved motif compositions.Analysis of cis-acting elements in the promoter region revealed that the promoter regions of GhSCL3 genes contained multiple elements related to abiotic stress response.Tissue expression pattern analysis showed that some GhSCL3 genes exhibited dominant expression in specific tissues.Expression analysis under drought treatment indicated that the expression of some GhSCL3 genes was significantly induced by drought stress,among which the expression of GhSCL3-D13-1 was significantly upregulated.qPCR further confirmed that the transcriptional level of GhSCL3-D13-1 could be significantly induced by PEG or ABA treatment.After silencing GhSCL3-D13-1 via VIGS,compared with the control plants,the leaf relative water content(RWC),catalase(CAT)and peroxidase(POD)activities of the gene-silenced plants were significantly decreased,while the malondialdehyde(MDA)content was significantly increased.These results indicated that GhSCL3-D13-1 positively regulated the drought stress response in cotton.关键词
