和元畅络生血方对肾性贫血大鼠的治疗作用及其机制
Therapeutic effect and mechanism of Heyuan Changluo Shengxue Formula on renal anemia in rats

Abstract
Objective:To explore the therapeutic effect and mechanism of Heyuan Changluo Shengxue Formula on re-nal anemia in rats with adenine-induced chronic renal failure(CRF).Methods:Thirty-three SD rats were randomly di-vided into a blank control group(9 rats)and an adenine modeling group(24 rats)according to the random number table method.The rats in the adenine modeling group were gavaged with adenine(200 mg/kg)for 4 weeks to induce a rat model of renal anemia.After successful modeling,the rats were randomly divided into a model group,a western medicine group and a traditional Chinese medicine(TCM)group,with 6 rats in each group.Rats in the western medicine group received recombi-nant human erythropoietin(rHuEPO)at 180 U/(kg·week);rats in the TCM group received Heyuan Changluo Shengxue Formula(3.0 g/(kg·d))by gavage in addition to the western medicine treatment;rats in the remaining groups received an equal volume of normal saline.All rats were treated for 8 weeks.After treatment,red blood cell(RBC)count,hemoglobin(Hb)level,hematocrit(HCT),serum urea nitrogen(BUN),creatinine(SCr)and erythropoietin(EPO)concentrations were measured in the four groups.Renal pathological changes were observed under hematoxylin-eosin(HE)staining.Results:HE staining results showed that renal pathological damage in the TCM and western medicine groups was signifi-cantly improved compared with the model group,and the recovery effect in the TCM group was superior to that in the west-ern medicine group.Compared with the blank control group,RBC,Hb,HCT and serum EPO levels in the model,western medicine and TCM groups were significantly decreased,while serum BUN and SCr levels were significantly increased.The differences were statistically significant(P<0.05).Compared with the model group,serum BUN and SCr levels in the west-ern medicine and TCM groups were significantly decreased,while RBC,Hb,HCT and serum EPO levels were significantly increased.The differences were statistically significant(P<0.05).Compared with the western medicine group,serum BUN and SCr levels in the TCM group were significantly decreased,while RBC,Hb,HCT and serum EPO levels were signifi-cantly increased.The differences were statistically significant(P<0.05).Conclusion:Heyuan Changluo Shengxue Formula can improve renal anemia in CRF rats by protecting renal function and increasing serum EPO concentration.关键词
