摘要

Abstract

Objective:To measure the differential mRNA expression profile of rheumatoid arthritis(RA)with blood stasis syndrome using transcriptome technology and perform biological information analysis to explore the intrinsic relation-ship between RA blood stasis syndrome and mRNA expression profile.Methods:A total of 5 patients each with RA blood stasis syndrome,RA non-blood stasis syndrome,normal people with blood stasis syndrome,and normal people without blood stasis syndrome were selected from Shenzhen Hospital of Huazhong University of Science and Technology Union Hospital from June 2022 to October 2022.Peripheral blood was collected,peripheral blood mononuclear cells were isolated,and total RNA was extracted for transcriptome sequencing.Biological information software was used to analyze the differential gene mRNA expression and enriched pathways in RA blood stasis syndrome and RA non-blood stasis syndrome patients.Reverse transcription polymerase chain reaction(RT-PCR)was used to verify the differentially expressed genes.Results:Transcrip-tome sequencing and bioinformatics analysis showed that compared with the RA non-blood stasis syndrome group,the RA blood stasis syndrome group had 32 differentially expressed mRNA genes(13 up-regulated and 19 down-regulated).Com-pared with the non-blood stasis syndrome group in normal people,there was 1 differentially expressed mRNA gene(1 up-regulated).Gene Ontology(GO)enrichment analysis of differential mRNA genes in the RA blood stasis syndrome group and RA non-blood stasis syndrome group showed enrichment mainly in 16 biological processes(BP),10 molecular functions(MF),and 7 cellular components(CC).Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis showed enrichment mainly in natural killer cell-mediated cytotoxicity(NKCMC),antigen presentation(AP),rheumatoid arthritis(RA),type Ⅰ diabetes(TID),and other signaling pathways.RT-PCR verification showed that compared with the RA non-blood stasis syndrome group,the mRNA expression of vascular endothelial growth factor A(VEGFA)and immune-related gene human leukocyte antigen-DQA1(HLA-DQA1)was up-regulated in the RA blood stasis syndrome group,with statistically significant differences(P<0.05).Conclusion:The differential mRNA genes in the RA blood stasis syndrome group and RA non-blood stasis syndrome group were mainly enriched in AP,RA,and NKCMC.RA can affect the expression of blood stasis syndrome,which is related to the up-regulation of VEGFA and HLA-DQA1 genes.
