苦荞转录因子FtNAC16在调控次生壁加厚中的功能研究
Functional validation of the transcription factor FtNAC16 in regulating secondary wall thickening in Tartary buckwheat

Abstract
NAC transcription factors play a crucial role in the thickening process of the secondary wall in plants.However,their functions in Tartary buckwheat(Fagopyrum tataricum)have been less investigated.To explore the function of FtNAC16,a candidate transcription factor identified in our previous study for secondary wall synthesis in buckwheat,Agrobacterium rhizogenes-mediated genetic transformation was employed to obtain transgenic buckwheat hairy roots.These roots were then analyzed using cytological examination,physiological measurements and gene expression analysis.The results showed that overexpression of FtNAC16 significantly increased the cell wall thickness and the contents of cellulose and hemicellulose in the transgenic buckwheat hairy roots.Furthermore,overexpression of FtNAC16 markedly upregulated the expression of multiple MYB transcription factors involved in secondary wall synthesis,as well as structural genes responsible for cell wall synthesis.These findings confirm that FtNAC16 functions in regulating the synthesis of secondary wall in buckwheat by controlling the expression of downstream MYB transcription factors and structural genes involved in secondary wall synthesis,thereby increasing the contents of cellulose and hemicellulose,and promoting cell wall thickness.This study provides valuable insights into the molecular mechanism of secondary wall synthesis in buckwheat and lays the theoretical foundation for improving the cell wall thickness in buckwheat pericarp.关键词
