基于circCOL1A1过表达/敲减的长江三角洲白山羊毛囊干细胞转录组测序及分析
Transcriptome Sequencing and Analysis of Hair Follicle Stem Cells with circCOL1A1 Overexpression and Knockdown in Yangtze River Delta White Goat

Abstract
This study aimed to investigate the differentially expressed genes(DEGs)in hair follicle stem cells(HFSCs)with circCOL1A1 overexpression or knockdown,and to preliminarily screen key genes regulating HFSCs growth and development in Yangtze River Delta White goat.HFSCs were isolated from the cervical spinal skin tissues of male Yangtze River Delta White Goat fetuses at 115-125 d of gestation.After overexpression or knockdown of circ-COL1A1,transcriptome sequencing and bioinformatics analysis were performed on the treated cells,followed by mul-tiple interaction networks that constructed by STRING and other databases.Finally,DEGs were further validated using real-time quantitative polymerase chain reaction(qPCR)and Sanger sequencing.The results showed that genes such as MYO16,HAPLN4,TBKBP1,CPXM1,TXINP CSRP2,and THEM5 were differentially expressed in HFSCs with knock-down of circCOL1A1,while genes including CMTM3,STAT1,MX1,SAMD9,and GBP5 were differentially expressed in HFSCs with overexpression of circCOL1A1.These genes participated in HFSCs growth and development through PI3K-Akt,RIG-Ⅰ-like receptor(RLR),JAK-STAT,and MAPK-Ras-ERK signaling pathways.This study found that differentially expressed genes such as MYO 16,HAPLN4,CMTM3,and STAT1 were mainly enriched in the PI3K-Akt and RLR pathways,and involved in regulating processes of growth and development of hair follicle stem cells.关键词
