Study on VIGS-Induced Gene Silencing in the DoDELLAs Family Gene of Dendrobium officinale

Abstract
To explore the function of DoDELLAs genes in Dendrobium officinale,the virus induced gene silencing(VIGS)technology,the pCymMV-Gateway recombinant vector targeting the DoDELLA1,DoDELLA2 and DoDELLA5 genes was constructed.The recombinant vector was introduced into Dendrobium officinale through the leaf injection method mediated by Agrobacterium to achieve effective silen-cing of the target DoDELLAs gene.RT-qPCR analysis confirmed that after silencing treatment,the expres-sion levels of DoDELLA 1 to DoDELLA 6 genes were significantly downregulated.Transcriptome sequencing identified 3 610 differentially expressed genes(DEGs),among which 1 670 were upregulated and 1 940 were downregulated.GO function and KEGG pathway enrichment analysis revealed that DEGs were respectively involved in key pathways such as plant hormone signal transduction,plant MAPK signaling pathway,and starch and sucrose metabolism.Studies had shown that silencing the DoDELLAs gene relieved its inhibition of the gibberellin signaling pathway,thereby activating the expression of genes related to promoting stem elon-gation and germination.Meanwhile,the suppression of the MAPK signaling pathway by DoDELLAs was lifted,activating the kinase signal cascade and inducing the expression of downstream defense-related genes,thereby enhancing the stress resistance of the plants.In addition,the reprogramming of energy metabolism pathways jointly drived a series of physiological changes,including promoting growth and development.关键词
