5个高粱品系染色体制片技术优化及核型分析
Optimization of Chromosome Preparation and Karyotype Analysis in 5 Strains of Sorghum bicolor

Abstract
To explore the karyotype characteristics of different strains of Sorghum bicolor,the enzy-matic hydrolysis titration method was used to conduct karyotype analysis on 5 strains,namely Jiniang No.1,Desheng Sweep Broom Sorghum,Jinnuo No.3,Jingdu Xiaobairen and Jinbai No.29.The results showed that mitotic metaphase chromosomes could be obtained by fixation w ith 90%glacial acetic acid and Carnoy solution respectively using the root tips of Sorghum bicolor as the material.After pretreatment with a mixture of ice and water and fixation with Carnoy solution,the chromo-some preparation effect was better,and the obtained chromosomes had clear morphology,moderate shrinkage and uniform dispersion.In this study,all 5 sorghum plants were diploid(2n=2x=20),all had karyotype 1A,and each had a pair of chromosome with satellite structures.The karyotype formu-la of Jiniang No.1 was 2n=2x=18m(2SAT)+2sm,that of Desheng Sweep Broom Sorghum was 2n=2x=2M+18m(2SAT),and the karyotype formulas of the other strains were all 2n=2x=20m(2SAT).The results of the karyotype asymmetry coefficient showed that their evolutionary degrees from high to low were as follows:Jinbai No.29,Jiniang No.1,Desheng Sweep Broom Sorghum,Jing-du Xiaobairen and Jinnuo No.3.关键词
