基于转录组测序探究营养对蝉花菌孢梗束发育的调控机制
Investigation of the regulatory mechanisms of nutritions on synnemata development of Cordyceps chanhua through transcriptomic sequencing

Abstract
In order to enrich the knowledge about the regulatory mechanism of synnemata development of Cordyceps chanhua,the regulatory mechanisms of nutritions were investigated at the transcriptomic level in this study.The results showed that Cordyceps chanhua cultured under lighting could form synnemata on wheat media,but could only form mycelia on yeast extract with supplements(YES)solid media.After culture for 28 d on the above two media,microscopic morphologies of Cordyceps chanhua showed significant differences during observation by cryo-scanning electron microscopy.The mycelia grown on YES solid media were made up of bouffant hyphae with apparent interspaces and abundant conidia.In contrast,the synnemata formed on wheat media showed as closely cohered bundles and no conidia were attached.Obviously different abilities of the above two kinds of media in inducing synnema formation suggested that nutritions play an important regulating role.After transcriptomic sequencing and analysis,2 484 differentially expressed genes(DEGs)of the fungus were obtained,which were caused by the above two kinds of media.Annotation results of DEGs'functions showed that the subclasses of gene ontology(GO)functions such as catalytic activity and cellular anatomical entity,and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathways such as biosynthesis of amino acids and carbon metabolism,enriched obviously abundant DEGs.The genes,such as inner centromere protein encoding gene and kynurenine 3-monooxygenase encoding gene,were strongly upregulated during culture with wheat media.On the contrary,the genes such as peptidase(A1)encoding gene and glucanase encoding gene,were strongly downregulated.The above results indicate that suitable nutritions are necessary for Cordyceps chanhua to form synnemata and that they regulate synnemata development through complex molecular mechanisms.关键词
