Abstract

This study aimed to identify the pathogenic fungi and investigate the associated mycoviruses responsible for peach gummosis in Henan province,China.Disease surveys were conducted in six peach orchards across the province,and a total of 31 fungal isolates were obtained using tissue isolation methods.Based on pathogenicity assays,morphological characteristics and molecular identification,the isolates were classified into four species:Botryosphaeria dothidea,Diplodia seriata,Lasiodiplodia theobromae,and L.citricola.Among them,L.citricola was recorded for the first time on peach trees in China,and B.dothidea was identified as the dominant pathogen.Detection of mycoviruses using dsRNA extraction and RT-PCR revealed that 11 B.dothidea strains contained dsRNA fragments ranging from 0.5 to 5 kb,and three mycoviruses[Botryosphaeria dothidea partitivirus(BdPV1),Botryosphaeria dothidea RNA virus 1(BdRV1),and Botryosphaeria dothidea chrysovirus 1(BdCV1)]were identified.Notably,BdRV1 significantly reduced the virulence of its fungal host,suggesting its potential as a biocontrol agent against peach gummosis.This study provides valuable theoretical and practical foundations for understanding the etiology and developing biological control strategies for peach gummosis disease.
