Abstract

Transgenic insect-resistant crops can effectively control target pests,however,with prolonged cultivation,the risk of pests developing resistance increases,thus,assessing pest resistance risk in advance and developing rational resistance management strategies are crucial for the sustainable application of transgenic crops.In this study,the sensitivity changes of Cry1Ac-resistant Helicoverpa armigera strains(LF10,LF30,LF60,LF120)feeding on three transgenic Bt maize cultivars('DBN9936''DBN3601T''Ruifeng 125')were determined using the diet incorporation bioassay.The transcriptional levels of major Bt receptor genes were compared between larvae fed on conventional maize and those fed on Bt maize using quantitative real-time PCR(RT-qPCR).The results indicated that the Cry1Ac-resistant strains exhibited reduced sensitivity to all three Bt maize cultivars compared with the susceptible strain,and the sensitivity declined with increasing Cry1Ac resistance level in the resistant strains.Feeding on Bt maize altered the expression of Bt receptor genes in H.armigera,with varying effects among maize cultivars and between different insect strains.Overall,the transcription of ABCC2 was down-regulated after fed on transgenic Bt maize.When feeding on the same Bt maize,resistant H.armigera showed significantly lower transcription levels of ABCC2 and CAD than the susceptible strain.These results suggest that ABCC2 may play a major role in the stress response of H.armigera under Bt protein exposure.The Cry1Ac-resistant H.armigera showed obvious cross-resistance to the three Bt maize cultivars(which express Cry1Ab proteins),and the significant down-regulation of ABCC2 and CAD in resistant strains may be the main molecular basis for the cross-resistance.
