摘要

Abstract

Objective To observe the effects of the sulfated homogeneous polysaccharide from codonopsis pilosula(SCOP-2)on the proliferation,apoptosis,and apoptosis-related genes of hepatocellular carcinoma HepG2 cells.Methods The MTT assay was used to determine the effect of SCOP-2 on the proliferation of HepG2 cells;Ho-echst 33258 staining(observed under a fluorescence microscope)and flow cytometry were applied to detect the effect of SCOP-2 on the apoptosis of HepG2 cells;Reverse transcription-quantitative polymerase chain reaction(RT-qPCR)and Western blot were further used to examine the effect of SCOP-2 on apoptosis-related gene/pro-teins(BCL-2,BAX,p53,and Caspase-3)in HepG2 cells.Results The experimental results showed that SCOP-2 could inhibit the proliferation and promote the apoptosis of HepG2 cells.The possible mechanisms for SCOP-2 to act on HepG2 cells appeared to be due to the down-regulation of BCL-2 expression and the up-regula-tion of BAX expression in the cells,which further promoting the release of the apoptosis-related factor p53.Sub-sequently,the caspase cascade reaction is activated,which promotes the expression and activation of Caspase-3,thereby facilitating the apoptosis of HepG2 cells.Conclusion SCOP-2 promotes the apoptosis of HepG2 cells by regulating the expressions of BCL-2,BAX,p53,and Caspase-3.
