摘要

Abstract

Using 15 pairs of self-designed and screened universal EST-SSR primers for Ulmus,we analyzed the genetic diversity and population structure of 184 Ulmus germplasm resources,including 6 species and 2 varieties.Additionally,we con-structed the core germplasm of Ulmus pumila with Nei's genetic distance among individuals as the primary control condi-tion.The results showed that the observed heterozygosity,expected heterozygosity,and unbiased expected heterozygosity of Ulmus germplasm resources were 0.439,0.590,and 0.591,respectively,with a Shannon's information index of 1.278,indicating rich genetic diversity.The 184 germplasm resources could be divided into 2 groups:GroupⅠmainly consisted of U.pumila germplasm,while most other Ulmus germplasm(including U.davidiana,U.lamellosa,U.parvifolia,U.macro-carpa,U.davidiana var.japonica,and U.laciniata,etc.)belonged to GroupⅡ.PCoA analysis and clustering results further verified the reliability of population division.Considering Nei's genetic distance among individuals,allele retention rate,and number of individuals comprehensively,the constructed core germplasm of 15 U.pumila individuals retained 92.4%of the al-leles from the original germplasm.The number of individuals,Shannon's information index,observed heterozygosity,expec-ted heterozygosity,and unbiased expected heterozygosity increased to 123.3%,119.3%,113.0%,113.7%,and 117.1%of the original population,respectively,indicating high representativeness.
