珍稀兰科植物掌裂兰的非共生萌发研究
Study on the asymbiotic germination of rare Orchidaceae plant Dactylorhiza hatagirea

Abstract
[Objective]Dactylorhiza hatagirea is an orchid plant used both as medicine and food in Linzhi City,Tibet Autonomous Region.To explore the effects of different influencing factors on the asymbiotic germination of Dactylorhiza hatagirea seeds.[Method]This experiment took the asymbiotic germination rate of Dactylorhiza hatagirea seeds as an indicator to investigate the effects of different basic media(MS,N6,and Hyponex No.1),different concentrations of exogenous hormones(NAA and 6-BA),different seed development times,and different concentrations of activated carbon and vitamins on the germination of Dactylorhiza hatagirea seeds.[Result]The results showed that:1.In terms of basic media,there was little difference in the germination rate of Dactylorhiza hatagirea seeds among MS,N6,and Hyponex No.1 basic media.2.Regarding single exogenous hormones at different concentrations,the germination rate of Dactylorhiza hatagirea seeds with the addition of NAA and 6-BA at any concentration was lower than that without the addition of exogenous hormones.3.In terms of seed development time,the germination rate of Dactylorhiza hatagirea seeds first increased and then decreased with the increase of development time.4.With respect to different additives,the germination rate of Dactylorhiza hatagirea seeds without the addition of activated carbon or vitamin C was 0%.The addition of activated carbon and vitamin C at different concentrations could improve the germination rate,but there were certain differences in the subsequent growth and development of protocorms.[Conclusion]The three basic media,namely 1,MS,N6,and Huabao No.1,have little impact on the seed germination of Dactylorhiza hatagirea.The exogenous hormones NAA and 6-BA have an inhibitory effect on the asymbiotic germination of Dactylorhiza hatagirea seeds.The germination rate of Dactylorhiza hatagirea seeds 60 days after pollination is the highest,while the seeds 30 days after pollination have no germination ability.2 g/L activated carbon and 0.5 g/L vitamins have the strongest stimulating effect on the embryos of Dactylorhiza hatagirea seeds whose activity has decreased after being stored in a refrigerator at-25℃for 6 months,resulting in the highest germination rate.The protocorms germinated from Dactylorhiza hatagirea seeds with activated carbon added develop better than those with vitamins added.关键词
