Establishment and evaluation of an in vitro regeneration system for Sarcopyramis nepalensis

Abstract
[Objective]This study is intended to explore a tissue culture and regeneration system for Sarcopyramis nepalensis and provide technical support for its large-scale seedling production.[Method]Using healthy,disease-free stem segments from robustly growing plants as explants,the suitable disinfection protocols were screened,and the effects of different basic media on adventitious bud induction,proliferation,and rooting were explored.Additionally,the influences of different transplanting substrates,seedling ages,and seedling hardening durations on the transplant survival rate of Sarcopyramis nepalensis were investigated.[Result]The best effect is achieved by disinfecting the explants with 0.1%HgCl2 for 11 minutes.The most suitable medium for adventitious bud induction is MS+2.0 mg/L 6-BA+0.1 mg/L NAA,with an induction rate of 84.4%.The optimal medium for proliferation is MS+0.3 mg/L 6-BA+0.1 mg/L NAA,with a multiplication factor of 6.12.The rooting culture effect is best when using 1/2MS as the basic medium anding 0.5 mg/L IBA+0.1 mg/L NAA+1.0 g/L activated charcoal,with a rooting rate of 100%.The mixture of peat soil and bark at a 3:1 ratio proved to be the optimal transplanting substrate,achieving a survival rate of 90.8%and a single-plant growth weight of 0.45 g.Transplanting the tissue-cultured seedlings at an age of approximately 60 days was found to be appropriate,as this stage yielded the highest transplant survival rate and the best growth performance.Seedling hardening for 7 days could better enhance the adaptability of the tissue-cultured seedlings after transplantation and significantly improve the transplant survival rate.[Conclusion]This study established a tissue culture and regeneration system for Sarcopyramis nepalensis.It provides technical guidance for the germplasm resource preservation and industrialized seedling production of Sarcopyramis nepalensis in the future,and also lays a foundation for its genetic transformation research.关键词
