摘要

Abstract

[Objective]To clarify the pathogen causing bacterial stem blight on winter tomato in Lianjiang County,Fujian Province,providing a theoretical basis for the control of this disease.[Method]The pathogen was isolated and purified using the plate streaking method.Its pathogenicity was confirmed through Koch's postulates.The pathogen's identity was determined by physiological and biochemical tests,as well as 16S rDNA gene sequence analysis.[Result]The results showed that seven bacterial strains were isolated and purified from diseased tomato stems,and Koch's postulates confirmed that all seven strains were the pathogens causing this disease.The bacterial cells were short rod-shaped and Gram-stain negative.Based on 22 physiological and biochemical tests,such as poly-β-hydroxybutyrate(PHB)production,yellow-green fluorescence production on King's B(KB)medium,fructan production,arginine hydrolysis,oxidase test,and utilization of glucose,xylose,cellobiose,maltose,lactose,trehalose,and glycine betaine et al,as well as 16S rDNA gene sequence analysis,the bacterial strains were identified as Pseudomonas viridiflava.[Conclusion]The tomato bacterial stem blight diseases in Fujian is caused by P.viridiflava.
