Abstract

Objective:Colorectal cancer is one of the leading causes of cancer-related mortality worldwide.This study aims to investigate the expression levels of kinesin family member C1(KIFC1)and family with sequence similarity 83 member A(FAM83A)in colorectal cancer tissues and to analyze the association between their expression and patient prognosis,in order to identify potential biomarkers for colorectal cancer. Methods:A total of 120 patients with colorectal cancer who were admitted to First People's Hospital of Kunshan between October 2017 and September 2019 were enrolled in this study.All patients were pathologically confirmed as stage I-III colorectal cancer and had not received preoperative radiotherapy or chemotherapy.Cancer tissues and adjacent non-cancerous tissues(>5 cm from the tumor margin)were collected during surgery.Immunohistochemical staining was performed to detect the protein expression of KIFC1 and FAM83A in both tissues.Expression status was determined based on staining intensity and the proportion of positive cells.Clinicopathological data,including gender,age,pathological stage,degree of differentiation,lymph node metastasis,and depth of invasion,were collected.All patients were followed up for 5 years after surgery,with colorectal cancer-related death as the endpoint.According to follow-up outcomes,patients were divided into a death group(n=31)and a survival group(n=89).Kaplan-Meier survival curves were used to analyze the relationship between KIFC1 and FAM83A expression and 5-year survival rate.Variables with statistical significance in univariate analysis were further included in a multivariate Cox proportional hazards regression model to calculate hazard ratio(HR)and 95%confidence interval(CI)and to identify independent prognostic factors. Results:Immunohistochemical staining showed that KIFC1 and FAM83A proteins were mainly localized in the nucleus.The positive expression rates of KIFC1 and FAM83A in colorectal cancer tissues were 60.83%(73/120)and 59.17%(71/120),respectively,both of which were significantly higher than those in adjacent non-cancerous tissues(both P<0.001).Prognostic analysis revealed that the positive expression rates of KIFC1 and FAM83A in cancer tissues from the death group(both 77.42%)were significantly higher than those in the survival group(55.06%and 52.81%,respectively;both P<0.05).Kaplan-Meier survival analysis demonstrated that the 5-year survival rate of patients with positive KIFC1 expression was significantly lower than that of patients with negative expression[67.12%(49/73)vs 85.11%(40/47);P<0.05].Similarly,patients with positive FAM83A expression had a significantly lower 5-year survival rate compared with those with negative expression[66.20%(47/71)vs 85.71%(42/49);P<0.05].Univariate analysis indicated significant differences between the death group and survival group in pathological stage,degree of differentiation,and depth of invasion(all P<0.05).Multivariate Cox regression analysis further confirmed that pathological stage III(HR=4.624,95%CI 1.964 to 10.899),poor differentiation(HR=5.108,95%CI 2.049 to 12.733),invasion depth T3 to T4(HR=5.429,95%CI 1.994 to 14.778),positive KIFC1 expression(HR=6.052,95%CI 2.112 to 17.338),and positive FAM83A expression(HR=4.927,95%CI 2.020 to 12.019)were independent risk factors affecting the prognosis of patients with colorectal cancer(all P<0.05). Conclusion:KIFC1 and FAM83A are highly expressed in colorectal cancer tissues and are associated with patient prognosis,suggesting that they may serve as potential biomarkers for prognostic evaluation in colorectal cancer.
