MS-222和丁香酚麻醉胁迫下四指马鲅幼鱼鳃转录组学分析
Transcriptomic analysis of gills in juvenile Eleutheronema tetradactylum under anesthetic stress

Abstract
Eleutheronema tetradactylum is mainly distributed in the Indian Ocean and western Pacific Ocean,and also in the East China Sea,South China Sea,and other waters.In recent years,its artificial breeding technology has made breakthroughs and has been promoted in coastal areas such as Guangdong,Guangxi,Fujian,Hainan,and Zhejiang provinces.It has now developed into a new variety of aquaculture,with high economic value and development prospects.At present,systematic research has also been conducted on the embryonic development,salinity tolerance,digestive tract structure,and functional cell development of E.tetradactylum.However,the oxygen exchange ability of E.tetradactylum is poor,and it is very sensitive to stress.To understand the effect of anaesthesia stress on juvenile E.tetradactylum,this study used MS-222(14 mg·L-1)and eugenol(0.1 mg·L-1)as anaesthetics to induce 12 hours of stress on juvenile E.tetradactylum.Transcriptomics analysis was used to investigate the differences in gene expression of juvenile E.tetradactylum under anaesthesia stress.After anaesthesia with eugenol(T1 group)and MS-222(T2 group),896 and 37 differentially expressed genes(DEGs)were identified in the gill tissues of juveniles,respectively.T1 group of DEGs was associated with 327 known KEGG pathways,while T2 group was associated with 153 known KEGG pathways.T1 differentially expressed genes enriched the intestinal immune network for IgA production,starch and sucrose metabolism,glycerophospholipid metabolism,and various other signaling pathways.T2 group enriched differentially expressed genes in intestinal immune network for IgA production,amino acid metabolism and various other signaling pathways.Long-term anesthesia with eugenol could cause inflammation in juvenile E.tetradactylum,while prolonged anesthesia with MS-222 could alter the osmotic regulation of juveniles.Eugenol anesthesia caused stronger stress on juveniles,and compared to eugenol anesthesia,MS-222 was more suitable for use as anesthesia for juvenile E.tetradactylum.After prolonged anesthesia with two types of anesthetics,the anesthetic stress on juvenile fish remained within a controllable range(no mortality was observed in juveniles after the experiment and 24 hours after resuscitation).关键词
