Abstract

Carotenoid cleaving oxidase(CCO)plays an important role in abiotic stress in plants as a key enzyme involved in the abscisic acid biosynthesis.To explore the function of the CCO gene family in grape,bioinformatics tools were employed to identify its members from the complete grape genome.Transcriptomic data were also analyzed to investigate the expression patterns of these genes across different grape tissues,during floral development and under adversity stress.The expressions were further validated by quantitative real-time PCR(qRT-PCR).A total of 14 VvCCO genes were identified and classified into six subfamilies based on phylogenetic analysis:CCD1,CCD4,CCD7,CCD8,CCD-like and NCED.Gene structures and conserved motifs exhibted similar distribution patterns within the same subfamilies.Analysis of cis-acting elements revealed that the largest proportion was associated with light response elements,followed by those related to phytohormones,environmental stresses,and plant developmental processes.Gene expression profiles showed that tissue-specific expression patterns of VvCCO genes.Among them,VvCCD1 gene showed the highest expression levels across all stages of flower development in different grapes.VvCCO gene family members showed no significant expression changes under salt stress while significant changes in expression levels under drought stress.qRT-PCR showed a significant increase in the expression of VvNCED1,VvNCED2,VvCCD4-1 and VvCCD4-2,and a significant decrease in the expression of VvCCD4-3 and VvCCD7 in Cabernet Sauvignon after two weeks of drought stress,which was in agreement with the reference transcriptome data(SRP182950).These results provide a theoretical foundation for further exploration into the function of CCO genes in grapes as well as their potential applications in breeding programs.
