益气健脾中药复方对糖尿病胃轻瘫大鼠Ghrelin的影响
Effect of Yiqi Jianpi TCM Compound on Ghrelin in Diabetic Gastroparesis Rats

Abstract
Objective Through the study of Ghrelin and its receptor GHSR in the gastric antrum tissue of DGP rats,to expore the the possibile mechanism on gastrointestinal motility in diabetic gastroparesis(DGP)rats.Methods The DGP rats were prepared by intraperitoneal injection of streptozotocin in combination with an irregular feeding method using a high-fat and high-sugar diet.After the successful establishment of the DGP model,the DGP rats were randomly divided into a Western medicine group(domperidone group),a traditional Chinese medicine group(Yiqi Jianpi TCM compound group),and a model group.In addition,six healthy Sprague-Dawley(SD)rats were set as the blank control group.After six weeks of gavage administration for treatment,the gastric emptying rate and small intestine propulsion rate of the rats in each group were determined.To observe the pathological changes of the gastric antrum tissue through HE staining.And determin the expression of Ghrelin,and measuring the expressions of Ghrelin and GHSR proteins by Western Blot(WB).Results Compared with the blank group,the fasting blood glucose of the rats in the model group increased(P<0.01),and the gastric emptying rate and small intestine propulsion rate were significantly slowed down(P<0.01).Through HE staining,in the mucosal layer of the gastric antrum tissue,the tissue arrangement was observed to be sparse,disordered and irregular,and the number of glands was significantly reduced.Compared with the model group:Compared with the model group,the fasting blood glucose level of the rats in the traditional Chinese medicine group was significantly decreased,and the difference was statistically significant(P<0.05).The gastric emptying rate and small intestine propulsion rate of the rats in both the traditional Chinese medicine group and the Western medicine group were significantly increased compared with those in the model group(P<0.05).HE staining showed that both the traditional Chinese medicine group and the Western medicine group improved the pathological state of the rat gastric antrum tissue to varying degrees.The expressions of Ghrelin and GHSR proteins in the gastric antrum tissue of the traditional Chinese medicine group and the Western medicine group increased compared with those in the model group(P<0.05).Compared with the Western medicine group:The expressions of Ghrelin and GHSR proteins in the traditional Chinese medicine group were increased compared with those in the Western medicine group(P<0.05).Conclusion The mechanism by which Yiqi Jianpi TCM compound and strengthens the spleen improves the general condition of DGP rats,reduces their fasting blood glucose,increases their gastric emptying rate and small intestine propulsion rate,and relieves their gastrointestinal motility disorder may be related to promoting the expressions of Gherelin and GHSR proteins.关键词
