Abstract

Objective:To investigate the optimal technological conditions for the preparation of Zuogutong Drop Pills and to explore their effects on the pain threshold in rats with neuropathic pain.Methods:The coefficient of variation of pill weight,roundness,and disintegration time were used as evaluation indices.The Box-Behnken response surface methodology was applied to analyze factors affecting the drop pill forming process.Drop pills prepared under optimal conditions were investigated by X-ray diffraction and scan-ning electron microscopy(SEM)to study physical mixtures of polyethylene glycol(PEG)6000 and PEG 4000,blank drop pills,and Zuogutong Drop Pills.A chronic constriction injury(CCI)model of the sciatic nerve was established in rats.The paw withdrawal mechanical threshold(PWMT)was used to evaluate mechanical pain threshold,and the paw withdrawal thermal latency(PWTL)was used to evaluate thermal pain threshold.Pathological changes in sciatic nerve tissue were observed,and the expression of N-methyl-D-aspartate receptor 1(NMDAR1)protein in the spinal dorsal root ganglia was determined by immunohistochemistry.Re-sults:Using PEG 4000 and PEG 6000 as the matrix,liquid paraffin as the condensing agent,a dropping distance of 5 cm,a medici-nal liquid-to-matrix ratio of 1∶3,a matrix ratio of PEG 6000 to PEG 4000 of 1∶2,and a medicinal liquid temperature of 75 ℃,the prepared drop pills exhibited small weight variation and good overall quality.X-ray diffraction and SEM results showed that PEG 6000 and PEG 4000 were incorporated into the matrix in a finer state.Pharmacodynamic experiments showed that,compared with the model group,rats in the drop pill group had higher PWMT and PWTL values(P<0.01),reduced sciatic nerve lesion severity(P<0.01),and lower NMDAR1 protein expression(P<0.01).Conclusion:The preparation process of Zuogutong Drop Pills is simple and the quality is stable,providing a basis for industrial production.Zuogutong Drop Pills can increase the pain threshold in rats with neuropathic pain,and their analgesic mechanism may be related to the downregulation of NMDAR1 expression.
