Mechanisms of Qufu Shengxin Ointment in the Treatment of Chronic Wounds from the Perspective of Autophagy

Abstract

Objective:To explore the potential mechanisms of Qufu Shengxin Ointment in the treatment of chronic wounds(CW).Methods:Network pharmacology was used to analyze the common targets of the active components of the ointment,CW,and auto-phagy.Key signaling pathway factors with strong correlations were selected for animal experimental validation.A CW rat model was established,and wound healing was observed.Histopathological changes were examined using hematoxylin-eosin(HE)staining,and the expression levels of autophagy-related targets,including mammalian target of rapamycin(mTOR),hypoxia-inducible factor-1α(HIF-1α),and vascular endothelial growth factor(VEGF),were evaluated by immunohistochemistry and quantitative real-time PCR(qPCR).Results:Network pharmacology identified 40 core targets,including mTOR and HIF-1α.Enrichment analysis high-lighted the PI3K-AKT and HIF-1 signaling pathways,with mTOR closely associated with these pathways.HE staining showed that Qufu Shengxin Ointment reduced inflammatory cell infiltration and promoted angiogenesis.Immunohistochemistry results demonstra-ted that mTOR protein expression in the Qufu Shengxin Ointment group was significantly higher than in the Fujifu group at days 7 and 14 post-treatment(P<0.05).HIF-1α protein expression at day 7 was higher than in both the model and Fujifu groups,while VEGF protein expression at day 14 was significantly higher than in these groups(P<0.05).As revealed by qPCR results,mTOR mRNA levels at day 7 were significantly higher than in the model and Fujifu groups(P<0.05).HIF-1α mRNA at day 7 was also significantly higher(P<0.05).VEGF mRNA was significantly increased at day 7 compared to the model group,and at day 14,it was significantly higher than in both the model and Fujifu groups(P<0.05).Conclusion:The external application of Qufu Shengxin Ointment may promote angiogenesis and accelerate CW healing,possibly through the regulation of autophagy-related path-way factors mTOR,HIF-1α,and VEGF.
