经典名方温经汤基准样品关键质量属性研究
Critical Quality Attributes of Wenjing Decoction Benchmark Sample

Abstract
Objective:To elucidate the critical quality attributes(CQAs)of the benchmark sample of Wenjing Decoction and pro-vide a reference for its development and quality control.Methods:High-performance liquid chromatography(HPLC)was employed to generate fingerprint profiles of 15 batches of Wenjing Decoction using the Similarity Evaluation System for Chromatographic Fin-gerprint of Traditional Chinese Medicine(2012 edition),and similarities were evaluated to identify common peaks.The extract yield,moisture content,extractable matter,and multiple component content of the benchmark sample were determined.Hierarchical cluster analysis(HC A)was performed using SPSS 21.0,while principal component analysis(PC A)and orthogonal partial least squares-discriminant analysis(OPLS-DA)were conducted using SIMCA 14.0 to assess quality differences among the 15 batches.Variable importance in projection(VIP)values were used to identify key marker compounds influencing quality.Results:A total of 20 common peaks were identified across the 15 batches of Wenjing Decoction.The extract yield ranged from 20.76%to 24.27%;moisture content from 5.25%to 7.51%;and extractable matter from 48.10%to 53.99%.The content of major components(mg/g)was as follows:gallic acid(0.28 to 0.39),paeoniflorin(0.45 to 0.55),liquiritin(0.18 to 1.13),ferulic acid(0.05 to 0.34),cinnamic acid(0.02 to 0.04),cinnamaldehyde(0.07 to 0.22),paeonol(0.41 to 0.61),ammonium glycyrrhizinate(1.31 to 2.72),ginsenoside Rg1(0.80 to 1.18),ginsenoside Re(0.51 to 0.76),and ginsenoside Rb1(0.90 to 1.55).Conclu-sion:The study comprehensively characterized the CQAs of Wenjing Decoction,providing a scientific basis for its formulation devel-opment and quality control.关键词
