柴胡-浙贝母药对及其活性成分治疗乳腺癌癌前病变大鼠的作用机制
Therapeutic Effects of Bupleuri Radix-Fritillariae Thunbergii Bulbus Herbal Pair and Its Active Components on Preneoplastic Breast Cancer in Rats

Abstract
Objective:To investigate the effects and mechanisms of the Bupleuri Radix-Fritillariae Thunbergii Bulbus herbal pair and its active components on preneoplastic breast cancer(PBC)in rats,focusing on angiogenesis mediated by the signal transducer and activator of transcription 3(STAT3)/vascular endothelial growth factor(VEGF)signaling pathway.Methods:Forty-five female Sprague Dawley(SD)rats were randomly assigned to five groups using a random number table:blank control,model,tamoxifen(TAM),Bupleuri Radix-Fritillariae Thunbergii Bulbus granules(granules),and saikosaponin D-peimine(monomer).Except for the blank group,the PBC model was established in the other groups by sequential administration of 7,12-dimethylbenz[a]anthracene(DMBA),estrogen,and progesterone.After successful modeling,the corresponding drugs were administered for 4 weeks.Therapeu-tic effects were evaluated by uterine and ovarian coefficients,hematoxylin-eosin(HE)staining,immunohistochemistry,quantitative real-time polymerase chain reaction(qPCR),and Western blot(WB)analyses.Results:Compared with the blank group,the model group showed severe ductal lesions,epithelial cell stacking,unclear ductal structures,and pronounced hyperplasia,confirming suc-cessful modeling.Compared with the model group,TAM,granules,and monomer groups all showed improvement in these pathologi-cal features,with reductions in uterine and ovarian coefficients and levels of VEGF-A,STAT3,basic fibroblast growth factor(bF-GF),and interleukin-6(IL-6)(P<0.01,P<0.05).Additionally,TAM and granules groups exhibited decreased microvessel den-sity(CD31-MVD)and phosphorylated STAT3(p-STAT3)levels(P<0.01,P<0.05).Conclusion:The Bupleuri Radix-Fritillari-ae Thunbergii Bulbus herbal pair and its active components may exert therapeutic effects on PBC by inhibiting STAT3/VEGF path-way-mediated angiogenesis.关键词
