Effects of Electroacupuncture on Intestinal Flora and P38 MAPK Pathway in Hypertensive Rats

Abstract
Objective:To investigate the effects of electroacupuncture on blood pressure,intestinal flora,ghrelin,and the p38 mito-gen-activated protein kinase/endothelial nitric oxide synthase(p38 MAPK/eNOS)signaling pathway in spontaneously hypertensive rats(SHRs),aiming to elucidate the antihypertensive mechanism of electroacupuncture and provide a scientific basis for its use in treating hypertension.Methods:Sixteen SHRs were randomly divided into a model group and an electroacupuncture group using a random number table,and eight Wistar-Kyoto(WKY)rats were included as a blank group.Rats in the electroacupuncture group re-ceived electroacupuncture(continuous wave,2 Hz,1 to 2 mA)for 20 min per day,5 times per week,for 4 weeks.Rats in the blank and model groups were grasped and immobilized for 20 min at the same time each day,5 times per week,for 4 weeks.Systolic blood pressure(SBP)and diastolic blood pressure(DBP)were measured using the tail-cuff method before and after the intervention.Fe-cal microbial communities were analyzed by 16S rDNA sequencing.Serum levels of ghrelin,thromboxane A2(TXA2),and nitric ox-ide(NO)were measured using enzyme-linked immunosorbent assay(ELISA).The expression of p38 MAPK and eNOS in vascular tissues was detected using quantitative real-time polymerase chain reaction(qRT-PCR).Results:Electroacupuncture significantly improved SBP and DBP in SHRs(both P<0.01).Compared with the blank group,the number of operational taxonomic units(OTUs)decreased in the model group;compared with the model group,the number of OTUs increased significantly in the electroa-cupuncture group(P<0.05),and the Shannon index was upregulated(P<0.05).Linear discriminant analysis effect size(LEfSe)showed that Lachnospiraceae_NK4A136_group,Ruminococcaceae_UCG_013,and Clostridiales were significantly enriched in the electroacupuncture group,whereas Allobaculum was significantly enriched in the model group.Correlation analysis indicated that Akkermansia(R=-0.592,P=0.02)and Clostridium_sensu_stricto_1(R=-0.515,P=0.049)were negatively correlated with DBP,and Prevotellaceae_UCG-001 was positively correlated with DBP(R=0.531,P=0.042).In addition,electroacupuncture in-creased serum ghrelin and NO levels and decreased TXA2 levels(both P<0.01).There was a trend toward decreased p38 MAPK expression and increased eNOS expression in the electroacupuncture group.Conclusion:Electroacupuncture effectively reduced SBP and DBP in SHRs,increased OTU numbers and microbial diversity in the intestinal flora,upregulated ghrelin,eNOS,and NO levels,decreased TXA2 levels,and downregulated p38 MAPK expression.The antihypertensive effect of electroacupuncture may be related to regulation of intestinal flora,ghrelin,and the p38 MAPK/eNOS signaling pathway.关键词
