The Relieving Effect of Sea Buckthorn and Tiger Nut Composite Beverage on Constipation Induced by Loperamide Hydrochloride in Mice

Abstract
In order to investigate the relieving effect of a compound drink containing sea buckthorn and tiger nut composite beverage(STB)on loperamide hydrochloride-induced constipation in mice.A constipation model was established by administering loperamide hydrochloride,and the mice were randomly divided into 4 groups:the negative control group(gavage sterilized water 0.2 mL/d per mouse),the model group(gavage 10 mg/kg lop-eramide hydrochloride 0.2 mL/d per mouse),the low-dose STB group(gavage 10 mg/kg loperamide hydrochlo-ride 0.2 mL/d+20.0 mL/kg STB 0.2 mL/d per mouse),and the high-dose STB group(gavage 10 mg/kg lop-eramide hydrochloride 0.2 mL/d+40.0 mL/kg STB 0.2 mL/d per mouse).The intervention effect of STB was evaluated by measuring body weight,intestinal propulsion rate,serum gastrointestinal neurotransmitter lev-els,intestinal tissue morphology,and intestinal flora structure.The results showed that compared with the control group,the model group exhibited significant(P<0.05)constipation symptoms,indicating successful modeling.Compared with the model group,the low-dose STB and high-dose STB groups showed a significant increase in fecal water content(65%and 60%,P<0.05),an increase in intestinal propulsion rate(78%and 75%,P<0.05),a reduction in the time to first black stool excretion(50%and 38%,P<0.05),and effective regulation of gastrointestinal neurotransmitter balance.Histopathological analysis revealed that STB improved the structure of colonic crypts and reduced inflammatory infiltration compared with the model group.16S rRNA sequencing analysis showed that after the STB intervention,the abundance of Firmicutes in the intestinal flora decreased,while the abundance of Bacteroidetes and beneficial bacterial genera increased significantly,and the flora structure tended to be healthier compared with the model group.In conclusion,STB alleviated constipation by promoting intestinal motility,regulating neurotransmitters,and restoring flora balance,providing a theoretical basis for the development of functional foods.关键词
