灵芝多糖对急性高眼压大鼠视神经损伤的抗氧化应激作用研究
Anti-oxidative stress effect of Ganoderma lucidum polysaccharides in acute high intraocular pressure-induced optic nerve injury in rats

Abstract
Objective To investigate the anti-oxidative stress effect of Ganoderma lucidum polysaccharides(GLP)in acute high intraocular pressure(IOP)-induced optic nerve injury in rats.Methods Forty-five female SD rats were ran-domly divided into a blank control group,a model group,and GLP groups(low-,medium-,and high-dose groups),with 9 rats in each group.Except for the blank control group,acute high IOP models were established in the right eyes of rats in the other groups by continuous anterior chamber perfusion with normal saline for 60 minutes.IOP was recorded before modeling,immediately after modeling,and 30 minutes after modeling.Rats in the low-,medium-,and high-dose GLP groups were administered GLP solution at 35,70,and 140 mg·kg-1·d-1,respectively,by gavage,while rats in the blank control and model groups received the same volume of normal saline by gavage.After 7 days of intervention,rats were sacrificed.Retinal morphology was observed and retinal thickness was measured by hematoxylin-eosin staining.Retinal cell apoptosis was detected by the TUNEL assay.Superoxide dismutase(SOD)activity was assessed by the xanthine oxidase method,and malondialdehyde(MDA)level was determined by the thiobarbituric acid method.Results Compared with the blank con-trol group,the model group showed a significant reduction in retinal ganglion cell(RGC)count and a decrease in average retinal thickness(both P<0.001).Compared with the model group,the medium-dose GLP group exhibited increased RGC count and thickened retina(both P<0.05),while the low-and high-dose GLP groups showed no statistically significant differences in average retinal thickness(both P>0.05).Compared with the blank control group,the model group had a sig-nificant increase in retinal apoptotic cell count(P<0.01).Compared with the model group,the low-,medium-,and high-dose GLP groups all showed reduced apoptotic cell counts in the retina(all P<0.05).Compared with the blank control group,the model group showed significantly decreased SOD activity and increased MDA level in the retina(both P<0.01).Compared with the model group,the medium-and high-dose GLP groups showed increased SOD activity and decreased MDA level in the retina(all P<0.05),while the low-dose GLP group showed no statistically significant differences(both P>0.05).Conclusion GLP mitigates oxidative stress injury by upregulating retinal SOD activity and downregulating MDA level,thereby inhibiting acute high IOP-induced RGC apoptosis.关键词
