Investigating the therapeutic effects and mechanisms of syringaresinol on knee osteoarthritis via the TLR4/NF-κB signaling pathway

摘要

Abstract

This study investigated the chondroprotective effects of syringaresinol(Syr)on rats with knee osteoarthritis(KOA)and its regulatory mechanism via the TLR4/NF-κB pathway.A KOA model was established in SD rats using the modified Hulth method.Thirty rats were randomly divided into a sham-operated group,a model group,a positive drug group(indomethacin,10 mg/kg),and low-and high-dose Syr groups(25 and 50 mg/kg),with six rats in each group.The results showed that compared with the model group,Syr intervention significantly improved motor function(P<0.01),reduced serum levels of IL-1β and MMP13(P<0.01),and alleviated cartilage pathological damage.Western blot and real-time quantitative polymerase chain reaction confirmed that Syr dose-dependently downregulated the protein and mRNA expression of TLR4 and P-NF-κB in cartilage tissue(P<0.01),while significantly increasing the expression of type Ⅱ collagen(P<0.01).Mechanistic studies indicated that Syr effectively alleviated joint inflammation and delayed cartilage matrix degradation by inhibiting the activation of the TLR4/NF-κB signaling pathway.These findings provide a theoretical basis for the clinical application of syringaresinol in the prevention and treatment of KOA and for the modernization of Chinese medicine.
