Abstract
To investigate the effects and underlying mechanisms of combined electromagnetic field exposure on neurobehavior and astrocytes in mice,C57BL/6N mice were randomly divided into four groups:Control group,Microwave group,Static magnetic field group,and Combined group(microwave+static magnetic field).An combined electromagnetic field exposure animal model was established.Neurobehavioral performance was evaluated using the Y-maze,tail suspension,and open field tests.Serum levels of aquaporin 4(AQP4),glial fibrillary acidic protein(GFAP),and CXC motif chemokine ligand 10(CXCL10)were measured by enzyme-linked immunosorbent assay(ELISA),while serum S100 calcium-binding protein β(S100β)was measured by radioimmunoassay.Expression of N-myc downstream-regulated gene 2(NDRG2)and GFAP in astrocytes of the hippocampal CA1 region was detected by immunofluorescence.The results showed that,compared with the Control group,the spatial exploration ability of mice in the Combined group decreased significantly on day 7 post-exposure,without exhibiting depression-or anxiety-like behaviors.At 6 h,7 d,14 d,and 21 d after combined exposure,serum levels of GFAP,CXCL10,and S100β were significantly increased(P<0.01 or P<0.001),whereas AQP4 levels were significantly decreased(P<0.01 or P<0.001).Furthermore,the expression of NDRG2 in hippocampal CA1 astrocytes was significantly downregulated on day 7 post-exposure(P<0.001).These findings indicate that combined exposure to microwave and static magnetic fields impairs spatial exploration,alters serum levels of astrocyte-related factors,and downregulates NDRG2 expression in hippocampal astrocytes.This study provides evidence for the involvement of astrocytes in the neurobiological effects of electromagnetic radiation.关键词
