摘要

Abstract

Objective:To analyze the genetic diversity of Rabdosia amethystoides and its related species,so as to lay a foundation for the identification of Rabdosia amethystoides and its related species and the breeding of excellent varieties.Methods:Using ITS,psbA-tmH barcodes and SRAP molecular markers respectively,and taking full advantage of the complementary advantages of the conservation and polymorphism of different barcodes,the identification and genetic relationship analysis of 20 samples of Rabdosia amethystoides and its related species were conducted.Results:Both the ITS and psbA-tmH phylogenetic trees showed that the 8 populations of Rabdosia amethystoides clustered into one branch,the 4 populations of Isodon rubescens clustered into one branch,the 3 populations of Isodon ja-ponicus and the remaining 5 related species of Rabdosia amethystoides clustered into one branch.Polymorphism detection,genetic diver-sity study and cluster analysis of Rabdosia amethystoides and its related species were conducted using 12 pairs of SRAP primers.When the genetic similarity coefficient was 0.723,the SRAP cluster map showed that Rabdosia amethystoides and Isodon lophanthoides clus-tered into one branch,Isodon rubescens and Isodon inflexus clustered into one branch,and other Camellia genus samples clustered into one branch.Conclusion:The phylogenetic tree constructed based on ITS and psbA-trnH sequences and the SRAP molecular markers can effectively distinguish Rabdosia amethystoides from its related species of the same genus.This study has guiding significance for the i-dentification of Rabdosia amethystoides and its related species as well as the screening of germplasm resources.
