基因标志物评分预测关节镜下部分半月板切除术后骨关节炎风险
Gene marker score predicts the risk of osteoarthritis after arthroscopic partial meniscectomy

Abstract
Objective To explore the expression of plasma mRNA in patients with osteoarthritis(OA)after arthroscopic partial menis-cectomy(APM),and construct a gene marker scoring model to assess OA risk.Methods A total of 268 patients who underwent APM at our hospital between January 2019 and December 2021 were included in this study.Using 1∶1 propensity score matching,78 patients were allocated to the OA group and 78 to the control group.Plasma samples from six participants in each group were subjected to mRNA microarray analysis.The hub genes were identified using KEGG pathway and protein-protein interaction(PPI)network analyses,and their expression was validated using real-time PCR,Western blotting(WB),and ELISA.Multivariate logistic regression was used to analyze the OA risk and the predictive performance of the variables was evaluated using receiver operating characteristic(ROC)curves.Results The incidence of OA was 29.1%(78/268).mRNA analysis identified 171 differentially expressed genes that were functionally enriched in pathways,such as extracellular matrix organization and FoxO signaling.PPI network analysis identified seven hub genes including COL1A1,COL3A1,HLA-DRA,and PXDN.Multi-technique validation confirmed that these four genes were significantly upregulated in the OA group(P<0.05).A gene marker score constructed based on these genes independently predicted OA risk in the multivariate ana-lysis(P<0.05),with an area under the curve of 0.931(0.879-0.965).Conclusion COL1A1,COL3A1,HLA-DRA,and PXDN have been implicated in OA pathogenesis.A gene signature score constructed based on these genes effectively predicted the OA risk and aided in the identification of high-risk patients.关键词
