摘要

Abstract

Objective:To conduct molecular identification of Lespedeza formosa and its mixed products by using DNA barcoding technology,in order to ensure the stable and reliable quality of clinical medication.Methods:A total of 27 samples of Lespedeza formosa from different origins and their similar easily mixed varieties were collected.The gene sequences of ITS2,psbA-trnH,matK and rbcL were used as candidate sequences for identification.Through PCR amplification and sequencing,the sequencing success rates and sequence characteristics of different candidate sequences were compared and evaluated,and the variations and genetic distances within and be-tween species were calculated.The phylogenetic tree was constructed.Results:Except for the matK sequence,the sequencing success rates of ITS2,rbcL,psbA-trnH were 100%,81.5%,66.7%,respectively.The ITS2 amplified sequence had more differential variation sites than the rbcL sequence,and some base variations could be used as identification features.Conclusion:The ITS2 sequence can ef-fectively distinguish the Lespedeza formosa and its mixed products,and can be used as the preferred identification sequence for DNA barcoding.This result provides a reference basis for enriching and improving the technical means of species authenticity identification of Lespedeza formosa and formulating relevant medicinal material quality standards.
