灯盏花乙素通过调控cAMP/PKA/CREB信号通路促进3T3-L1细胞脂肪分解的作用研究
Effects of scutellarin on lipolysis of 3T3-L1 cells by regulating cAMP/PKA/CREB signaling pathway

Abstract
Objective:To explore the effect of scutellarin(SCU)on 3T3-L1 cell lipolysis and its possible molecular mecha-nism in vitro,and provide reference for the treatment of metabolic syndrome.Methods:After inducing 3T3-L1 cells to mature adi-pocytes,cell viability of 3T3-L1 treated with SCU was assessed by CCK-8 assay.Lipid accumulation level of the cells was ana-lyzed,and the contents of total cholesterol(TC)and triacylglycerol(TG)in the cells were determined.The contents of tumor ne-crosis factor-α(TNF-α),interleukin-1β(IL-1β),IL-4 and IL-6 in the supernatant of cell culture were evaluated by ELISA.Real-time fluorescence quantitative PCR was used to detect the mRNA of uncoupling protein-1(UCP-1),deiodinase gene 2(Dio2)and peroxisome proliferator-activated receptor γ coactivator 1α(Pgc-1α)and the expressions of CREB,HSL and ATGL proteins were determined by Western Blot.Results:SCU had no significant effect on the viability of adipocytes,but could signifi-cantly inhibit lipid accumulation in 3T3-L1 cells,decrease the contents of TC and TG in the cells.Meanwhile,SCU decreased the levels of TNF-α and IL-1β and increase the levels of IL-4 and IL-6 in the supernatant.The mRNA expression of UCP-1,Dio2 and Pgc-1α in the cells was also upregulated,while the expression of p-HSL,ATGL,p-CERB and p-PKA proteins was reduced(P<0.05).Conclusion:SCU can alleviate the inflammatory response of 3T3-L1 adipocytes,promote lipidolysis by inhibit TG synthesis and lipid accumulation,and its mechanism is related to the activation of cAMP/PKA/CERB signaling pathway,which provides a necessary reference for the exploration of anti-metabolic syndrome effects of SCU.关键词
