青海省蚕豆赤斑病牡丹葡萄孢病原菌分离鉴定、生物学特性及致病性分化研究
Study on Isolation,Identification,Biological Characteristics and Pathogenicity Differentiation of the Pathogen of Broad Bean Chocolate Spot in Qinghai Province

Abstract
To clarify the biological characteristics and pathogenicity of Botrytis paeoniae,the pathogen of broad bean red spot in Qing-hai Province,the target strain was isolated and purified from the lesion tissue by using a tissue separation method.The morphologi-cal observation,molecular biological identification and biological characteristics were used for analysis,and 12 broad bean resourc-es in Qinghai Province were selected for pathogenicity evaluation.Morphological characteristics revealed that the colony edge was regular,the sclerotium was irregularly distributed,and the mycelium was light brown,with a two-branched structure and a dia-phragm.The conidia were mostly oval to rod-shaped,and the conidiophores were round,light brown or nearly transparent.Molecu-lar identification based on ITS and G3PDH gene sequence analysis confirmed the pathogen as Botrytis paeoniae;Biological character-istics indicated that Botrytis paeoniae suffruticosa grew fastest on PDA medium.The optimum carbon source was glucose,and the op-timum nitrogen source was beef extract and potassium nitrate.The optimum pH was neutral(pH 7).Under light conditions,12 h light/12 h dark or 24 h continuous light was more conducive to growth.Pathogenicity differentiation showed that different faba bean resources had different sensitivities to pathogens,and the pathogenic effect on'Yun 122'was the most significant.关键词
