Abstract

[Objective]The paper aimed to clarify the specific function of the EuNF-YC2 gene in order to create new stress-resistant varieties of Eucommia ulmoides.[Method]Based on the complete coding sequence of EuNF-YC2,the gene was cloned using specific primers,and bioinformatics analysis,subcellular localization,transcriptional activation,expression patterns,and yeast heterologous expression analysis were performed on the gene.[Result]The coding region of EuNF-YC2 was 792 bp in length,encoding 264 amino acids,with a molecular weight of 29.38 ku.The encoded protein was an acidic,hydrophilic,unstable protein containing a conserved HFD_NFYC domain at the N-terminus.Phylogenetic analysis indicated that the EuNF-YC2 protein was closely related to the tea plant NF-YC2.Subcellular localization analysis indi-cated that the EuNF-YC2 protein was localized to the nucleus,and both yeast and dual luciferase assays confirmed its transcriptional activa-tion activity.qRT-PCR results showed that the EuNF-YC2 gene exhibited a sustained upregulation pattern under salt stress,with a response intensity significantly higher than that under drought,high temperature and low temperature treatments.Additionally,overexpression of the EuNF-YC2 gene significantly enhances yeast cells'tolerance to salt stress and growth rate,and significantly improved the survival rate and chlorophyll content of transgenic Arabidopsis thaliana.[Conclusion]EuNF-YC2 is a key positive regulatory factor for E.ulmoides in response to salt stress.It is located in the nucleus and has transcriptional activation activity.Overexpression of the EuNF-YC2 gene can enhance the salt tolerance of plants and plays an important role in salt stress response,providing important genetic resources and theoretical basis for sub-sequent molecular breeding of E.ulmoides under stress resistance.
